چکیده
Introduction: Currently, short interfering RNA (siRNA) is an experimental tool in molecular biology to study gene function cell culture and vivo model of organisms (mouse chicken embryos, etc.) Method: Lentivirus production technique by human embryonic kidney 293T lines (HEK293T) with two (2) sites 8664/P201 2122/Pll.3.7 the sphingosine 1 phosphate receptor endothelial differentiation (S1P1/ Chicken EDG1) was used show effects small fibroblasts during embryogenesis. After preparation plasmids 2122/Pll3.7, lentivirus packaging, transfection 2xHBS, virus titration egg preparation, microinjection performed embryos chicken. Results: The results obtained after into from second day fourth incubation showed development materialized bleeding yolk sac therefore a decrease function. Conclusion: Previous studies have already proven that expression decreases certain vertebrates such as mammals.

Gene silencing of myostatin in differentiation of chicken embryonic myoblasts by small interfering RNA.
Myostatin (GDF-8) is known to negatively regulate skeletal muscle mass in myogenesis, but few studies have been conducted on the function of endogenous GDF-8 in primary myoblasts. The present study was performed to assess the function of GDF-8 by RNA interference using primary culture of chicken embryonic myoblasts in which myoblasts were differentiated into myotubes. An active form of small in...
متن کاملMyosin synthesis in embryonic chicken fibroblasts
The rate of constitutive myosin synthesis was measured in cultures of replicating embryonic chicken skin fibroblasts by pulse labeling with [3H]leucine. These cells synthesized the 200,000-dalton heavy chain of myosin (MHC) at a rate of 3.2 x 10(3) molecules/cell/min. Additionally, an independent estimate of the MHC synthesis rate needed to maintain a constant level of constitutive MHC/cell was...
متن کاملSmall interfering RNA; principles, applications and challenges--
Gene silencing using RNAi (RNA interference), has recently been used as a successful laboratory technique in determining the function and control of gene expression and provides a wide range of applications in molecular biology and gene therapy. RNAi is a method of suppressing gene expression. In this direction, a single-stranded RNA molecule of about 21–23 nucleotides, called siRNA (small inte...
متن کاملThe Effects of in ovo Nanocurcumin Administration on Oxidative Stress and Histology of Embryonic Chicken Heart
This study was designed to evaluate the effects of nanocurcumin (NC) on oxidative stress and histology of embryonic chicken heart. NC was injected into the yolk of 4-day-old embryonic eggs at one of three doses: 10 ppm (NC10 group), 100 ppm (NC100 group), and 1000 ppm (NC1000 group). The control group received normal saline. Oxidative stress in heart tissue was evaluated by measuring malondiald...
متن کاملDesign, simplified cloning, and in-silico analysis of multisite small interfering RNA-targeting cassettes
Multiple gene silencing is being required to target and tangle metabolic pathways in eukaryotes and researchers have to develop a subtle method for construction of RNA interference (RNAi) cassettes. Although, several vectors have been developed due to different screening and cloning strategies but still some potential limitations remain to be dissolved. Here, we worked out a simple cloning stra...
